CD163isexpressedspecificallyinthemonocyte/macrophagelineage,whereitmediatesuptakeofhaptoglobin‐hemoglobincomplexes,leadingtometabolismoftheoxidativehememolecule.SheddingoftheCD163ectodomainfromthecellsurfaceproducesasCD163plasmaprotein,andapositivecorrelationisseenbetweenthesCD163plasmalevelandtheseverityofvariousinfectiousandinflammatorydiseases.Inthepresentanalysisofthephorbolester‐inducedsheddingofsCD163inCD163cDNA‐transfectedHEK293cells,weusedmetalloproteinaseinhibitorsandsiRNA‐mediatedinhibitionofmetalloproteinasestoidentifyTACE/ADAM17asanenzymeresponsibleforPMA‐inducedcleavageofthemembrane‐proximalregionofCD163.AsTACE/ADAM17‐mediatedsheddingofTNF‐αisup‐regulatedinmacrophagessubjectedtoinflammatorystimuli,thepresentresultsnowprovidealikelyexplanationforthestrongempiricalrelationshipbetweenthesCD163plasmalevelandinfectious/inflammatorydiseasesrelatingtomacrophageactivity...
