OurprimaryaiminthisstudywastotestthehypothesisthatPEA,amemberofthefattyacidethanolamidefamilyandanendogenousPPAR‐αligand,exertsanti‐inflammatoryeffectsonSAOshock,causingasevereformofcirculatoryshockandenhancedformationofROS.SAOshockwasinducedbyclampingthesuperiormesentericarteryandtheceliactrunk,resultinginatotalocclusionofthesearteriesfor30min.Afterthisperiodofocclusion,theclampswereremoved.Inthisstudy,wedemonstratedthattheadministrationofPEA,5minbeforereperfusion,significantlyreducedalloftheparametersinvolvedduringinflammation,suchasproinflammatorycytokineproduction(TNF‐α,IL‐1β),adhesionmolecules(ICAM‐1,P‐selectin)expression,NF‐κBexpression,andapoptosis(Bax,Bcl‐2,TUNELassay)activation.Inaddition,tostudywhethertheprotectiveactionofPEAonSAOshockisalsorelatedtotheactivationofPPAR‐α,wehaveinvestigatedtheeffectofPEAinPPAR‐αKOmicesubjectedtoSAOshock.OurstudyclearlydemonstratesthatPEAsignificantlyattenuatedthedegreeofintestinalinjuryandinflammationcausedbyI/Rinjury.Moreover,thepositiveeffectsofPEAwereatleastinpartdependentonthePPAR‐αpathway.TheresultsclearlyindicatethatPEAexertsananti‐inflammatoryeffect,alsoinaSAOshockmodel,whichcouldimplyafutureuseofPEAinthetreatmentofI/Rshock...
