DeclineinCD4Tcellimmuneresponsesisassociatedwithaging.Althoughanumberofimmunologicaldefectshavebeenidentifiedinelderlymice(>18monthsold),akeyearly‐onsetimmunedefectatmiddleagecouldbeadriverorcontributortodefectiveCD4Tcellresponses.Ourstudiesdemonstratethatage‐relatedalterationsinDCsubsetswithintheprimingenvironmentofmiddle‐agedmice(12monthsold)correlatewithandcandirectlycontributetodecreasesinantigen‐specificCD4TcellTh1differentiation,whichmeasuredbyT‐betandIFN‐γexpression,wasdecreasedsignificantlyinTcellsfollowingVSVinfectionors.c.immunizationwithaproteinantigeninthecontextofimmunestimulationviaOX40.ThedeficientTh1phenotype,observedfollowingproteinantigenchallenge,wasfoundtobetheresultofanage‐relateddecreaseinaninflammatoryDCsubset(CD11b+Gr‐1/Ly6C+)inthedLNthatcorrespondedwithTcelldysfunction.Inthevirusmodel,weobservedsignificantchangesintwoDCsubsets:mDCsandpDCs.Thus,different,earlyage‐relatedchangesintheDCprofileintheprimingenvironmentcansignificantlycontributetoimpairedTh1differentiation,dependingonthetypeofimmunologicalchallenge...
