TheT‐boxtranscriptionfactorEomesisexpressedincytotoxicimmunecellsandplaysanimportantroleindevelopment,maintenance,andfunctionofthesecelltypes.IdentificationandseparationofcellswithdifferentialEomesexpressionwouldallowforbetterunderstandingofthetranscriptionalprogramgoverningthesecytotoxiclymphocytes.Here,wereporttheuseofanEomesgfp‐targetedmouseallelethatdisplaysrobustfidelitytoEomesproteinexpressioninNKandTcells.UseofthisreportermouserevealedthatEomesexpressioninantiviraleffectorcellsdidnotcorrelatewithenhancedcytotoxicitybutratherwasassociatedwithmoreefficientcentralmemorydifferentiation.WeakeningofreporteractivityinEomes‐deficientCD8+TcellsrevealedacriticalroleforEomesproteininmaintainingcentralmemorycellsthathaveactivatedtheEomeslocus.CharacterizationofreporteractivityinNKlineagecellsalsopermittedidentificationofanovelintermediateofNKcellmaturation.Thus,themurineEomesgfp‐targetedalleleprovidesanovelopportunitytoexploreEomesbiologyincytotoxiclymphocytes...
